Overview
ISO 12966-1:2014 provides guidelines for the gas chromatographic (GC) determination of fatty acids in animal and vegetable fats and oils after they have been converted to fatty acid methyl esters (FAMEs). Part 1 is a high-level guideline to modern gas chromatography of FAMEs, describing principles, column selection, detection choices and chromatogram evaluation. It complements Parts 2–4, which cover FAME preparation methods and detailed analytical conditions.

- Wall-coated open tubular (WCOT) capillaries are state-of-the-art. Column lengths typically range 10–100 m, internal diameters from 0.10–0.53 mm, and film thickness 0.1–0.3 μm.
- Highly polar cyanoalkyl polysiloxane phases (e.g., SP-2560, CP-Sil88) are recommended for separating geometrical/positional isomers and cis/trans isomers; other phases (BPX70, DB-23, Rtx-2330, etc.) may be used but can change elution order.
- Longer columns increase resolution (but with diminishing returns) and may be required for full cis/trans separation.
- Chromatogram evaluation
- Procedures for peak area, area percent, and use of internal standards or correction factors are included for quantitative work.
